2-Amino-3-methoxybenzonitrile was prepared by catalytic hydrogenation of 3-methoxy-2-nitrobenzonitrile, yielding a yield of 87.5%. The specific synthesis procedure is as follows: A solution of 3-methoxy-2-nitrobenzonitrile (Compound 9) (8.0 g, 44.9 mmol) in AcOEt (80 ml) was hydrogenated at atmospheric pressure using 10% Pd/C (800 mg) for 2 h. The catalyst was removed by filtration and the filtrate was concentrated. The residue was chromatographed on silica gel (CHCl3–MeOH, 30:1) to afford 2-amino-3-methoxybenzonitrile (5.9 g, 87.5%) as a colourless powder, melting point 64–67 °C[1].
