White crystalline powder, soluble in organic solvents such as methanol, ethanol, and DMSO, derived from Kava Piper methysticum.
anticonvulsant, analgesic, anxiolytic, suppreses excitatory postsynaptic potential
rac-Kavain is the main kavalactone found in the roots of the kava plant. Kavain is known to exhibit anticonvulsive properties by attenuating vascular smooth muscle contraction through interactions with voltage-dependent sodium and calcium channels. rac-Kavain was shown in recent research to exhibit protective properties on anaerobic glycolysis, ATP content and intracellular calcium and sodium of anoxic brain vesicles.
ChEBI: DL-kavain is https://www.chemicalbook.com/ProductChemicalPropertiesCB9738874_EN.htm It has a role as a glycine receptor antagonist.
A method for isolation and purification of Kavain, the method comprising the steps of:
(1) Pre-treatment of raw herbs: take Cava pepper and dry it at low temperature, and pulverize it into coarse powder;
(2) Supercritical extraction: put the crude powder of medicinal herbs into the extraction kettle for extraction, the extraction conditions are: pressure 18-30MPa, temperature 30-45 , time for 2-4 hours, and then the pressure is 4-8MPa, temperature 40-50 under the resolution;
(3) Column chromatography: take out the extract, heat water dispersion and add the macroporous adsorbent resin column adsorption, flow rate of 1-3BV/h, first 3 times the volume of the column 30% ethanol elution, and then 5 times the volume of the column of 50% -80% ethanol elution, collection of 50% -80% ethanol eluent;
(4) decolorization: the eluent is added with activated carbon, the amount added is 1-5% of the volume of the eluent;
(5) crystallization: decolorized liquid concentrated under reduced pressure precipitation crystal, crude crystal and then ethanol - ethyl acetate (1:1) heating to dissolve, refrigerated crystals, crystals drying drunken pepper pigment.
