Uses
Adamantanamine Hydrochloride is an antiviral that is used in the prophylactic or symptomatic treatment of influenza A. It is also used as an antiparkinsonian agent, to treat extrapyramidal reactions, and for postherpetic neuralgia. It is also used an NMDA-receptor antagoinst.
Synthesis
a kind of synthetic method of adamantanamine hydrochloride, comprises the steps:
1. react: 5 parts of (massfraction) sulfuric acid are joined in reaction vessel, with cold salt bath control temperature to-7 DEG C, drip 0.5 part of acetonitrile, again temperature is controlled at-3 DEG C, add 0.01 part of nickel catalyzator, then 1 part of amantadine is divided into two batches joins in above-mentioned mixed solution, after amantadine has added, be warming up to 18 DEG C, be incubated 6.5 hours;
2. be hydrolyzed: separately get a reaction vessel, add 28 parts of water, make its temperature be no more than 0 DEG C with ice bath, drip the liquid of step 1. after insulation, then add 13 parts of methylene dichloride wherein, stir 25 minutes;
3. extract: stopping is stirred and after stratification, the aqueous phase got wherein extracts with 6 parts of methylene dichloride;
4. dissolve: by the methylene dichloride evaporate to dryness after extraction, 7.5 parts of propyl carbinols are added in residue, 1 part of sodium hydroxide is added after stirring and dissolving, be heated to can carry out back flow reaction and react 7.5 hours, add 9.5 parts of water after cooling and stir 25 minutes, stop stirring treating its layering, get organic phase 4.5 parts of water washings once;
