7-Methoxy-6-(3-morpholin-4-ylpropoxy)quinazolin-4(3H)-one is a pharmaceutical intermediate used in the preparation of gefitinib, an orally active selective inhibitor of the epidermal growth factor receptor tyrosine kinase, an enzyme that regulates intracellular signalling pathways that associated with the proliferation and survival of cancer cells.
The preparation method comprises the following steps: with isovanillin as a raw material, carrying out cyanation and nitration so as to obtain 5-hydroxy-4-methoxy-2-nitrobenzonitrile; then carrying out one-pot reduction and cyclization so as to obtain 6-hydroxy-7-methoxy-(3H)-quinazoline-4-one; and finally, carrying out butt joint with a side chain so as to obtain 7-Methoxy-6-(3-morpholin-4-ylpropoxy)quinazolin-4(3H)-one.
