名称2-Thenoyltrifluoroacetone
描述2-Thenoyltrifluoroacetone (TTFA) is a specific inhibitor of mitochondrial respiratory chain complex II (SQR/SDH) with an IC₅₀ value of 51.5 μM. 2-Thenoyltrifluoroacetone can complex with various metal ions and exhibits cytotoxic and antitumor activity. It possesses anti-Mycobacterium tuberculosis activity and can inhibit the growth of Mycobacterium tuberculosis. 2-Thenoyltrifluoroacetone can be used in mechanistic studies of diseases associated with mitochondrial dysfunction.
体外活性方法：稳定转染KFERQ-PA-mCherry1的RTH-149虹鳟鱼肝细胞系加入2-Thenoyltrifluoroacetone 10 μM和高糖 25 mM，共处理16小时，荧光显微镜计数CMA puncta。 结果：2-Thenoyltrifluoroacetone 处理阻断了线粒体电子传递链复合物II的活性，阻断了高糖诱导的CMA puncta形成。[1] 方法：TET21N、SK-N-BE(2)、SH-SY5Y神经母细胞瘤细胞、U1810肺癌细胞加入 2-Thenoyltrifluoroacetone 100 μM和顺铂 5 μg/mL，共处理24小时，Western Blot检测cleaved caspase-3、cleaved PARP水平。 结果：常氧下，2-Thenoyltrifluoroacetone 显著增强顺铂诱导的凋亡，caspase-3和cleaved PARP 表达显著增加。[2] 方法：PC12大鼠嗜铬细胞瘤细胞加入2-Thenoyltrifluoroacetone 10 μM及鱼藤酮 1 μM，共处理24小时，H₂DCFDA荧光染色检测细胞内H₂O₂水平。 结果：2-Thenoyltrifluoroacetone 显著减少鱼藤酮诱导的H₂O₂产生。[3]
