名称	Palmitic acid
描述	Palmitic acid (Cetylic acid) is a natural product, a common saturated fatty acid found in animals, plants and microorganisms. Palmitic acid has antitumor activity.
激酶实验	As sources of PLA2, human recombinant sPLA2 (type IIA) is purified from CHO cells transfected with the PLA2 gene and rabbit recombinant platelet cPLA2 is obtained through its expression in baculovirus. The standard reaction mixture (200 μL) contained 100 mM Tris-HCl buffer (pH 9.0) with 6 mM CaCl2 and 20 nmol 1-acyl-[1-14C]-arachidonyl-sn-glycerophosphoethanolamine (2000 cpm/nmol) in the presence or absence of Tanshinone I. The reaction is started by adding 50 ng purified sPLA2 or cPLA2. After 20 min at 37°C, the free fatty acid generated is analysed. Under these standard conditions, the reaction mixture in the absence of Tanshinone I released approximately 10% of free fatty acid from the phospholipid substrate added[1].
体外活性	方法：小鼠小胶质细胞 BV2 用 Palmitic acid (25-200 nM) 处理 6-48 h，使用 MTT 方法检测细胞活力。结果：Palmitic acid 对小胶质细胞活力有浓度和时间依赖性抑制作用。[1] 方法：单核细胞衍生的树突状细胞 MoDC 用 Palmitic acid (150 μM) 处理 12 h，使用 Flow Cytometry 检测 DC 共刺激因子的表达。结果：Palmitic acid 诱导了 CD86 和 CD83 的表达，表明同时诱导了 MoDC 的激活和成熟。[2]
体内活性	方法：为研究对小鼠的不良生物行为的影响，将 Palmitic acid (0.3-30 μmol/mouse in castor oil) 单次腹腔注射给 C57BL/6J 小鼠，2-24 h 后对生物行为进行分析。结果：以剂量依赖的方式，Palmitic acid 通过依赖于脂肪酸链长度的机制快速降低小鼠运动活性。Palmitic acid 给药 24 h 后，小鼠表现出焦虑样行为，而运动、食物摄入、抑郁样行为或空间记忆没有受损。[3] 方法：为研究对心肌损伤的影响，将 Palmitic acid (5 mM in 500 μL solution) 尾静脉注射给 wild-type C57BL/6 (B6) 和 Md2-/- (KO) 小鼠，每天一次，持续七天。结果：Palmitic acid 通过 TLR4 辅助蛋白 MD2 诱导心肌炎症损伤，保护 Md2 敲除小鼠免受 Palmitic acid 和高脂肪饮食诱导的心肌损伤。[4]
