The general procedure for the synthesis of 2,6-dimethylbenzylamine from 2,6-dimethylbenzonitrile is as follows:
Example 1: The synthetic scheme for the preparation of 2-{2-[1-(2,6-dimethylbenzyl)-6-fluoro-1H-benzimidazol-2-yl]-5-methoxy-3-methylphenoxy}acetamide ("A1") is as follows:
1.1 2,6-dimethylbenzonitrile ("1") was dissolved in a mixed solution of methanol and 10% ammonia, and the hydrogenation reaction was carried out at 5 bar hydrogen pressure and 50°C for 16 hours using nickel Nguyenne as a catalyst. Upon completion of the reaction, the reaction mixture was filtered through diatomaceous earth and the solvent was subsequently removed under vacuum to give the product "2" as an oil in quantitative yield. Results of LC-MS analysis: retention time 1.307 min, m/e 136.5 (M+H+).
