4-Bromo-1-fluoro-2-nitrobenzene is used in the synthesis of anti-inflammatory agents.
4-Bromo-1-fluoro-2-nitrobenzene undergoes Sonogashira reaction with 2-fluoronitrobenzene to afford predominantly the bromo displacement product.
General procedure for the synthesis of 4-bromo-1-fluoro-2-nitrobenzene from 1-fluoro-2-nitrobenzene: After completion of the reaction, the reaction mixture was cooled to room temperature and slowly poured into ice water. The product 2-fluoro-5-bromonitrobenzene in the aqueous phase was extracted with 200 mL of ethyl acetate, and the organic phases were combined and concentrated under reduced pressure to give a brown solid crude product. The crude product was purified by recrystallization from petroleum ether and analyzed by gas chromatography (GC) with a purity of 98%, and 227.1 g of the pure product was obtained after drying with a yield of 40.0%.
