Methyl violet 2B is a biological stain that is commonly used at a pH between 0 and 1.6. It is certified for use in Flemming triple stain with iodine for chromosomes; Newton?s crystal violet-iodine technique for chromatin and nucleoli; Lieb?s amyloid stain.

Methyl violet 2B is a cationic dye with a triphenylmethane structure. It acts as a selective dye and facilitates the differentiation of Klebsiella pneumoniae from Enterobacter aerogenes and other gram-negative organisms. Methyl violet 2B is harmful if inhaled, swallowed or absorbed through skin, contact may cause damage to skin and eyes. Hence, the dye removal is of gre at importance.
Crystallise the dye from EtOH by precipitation with Et2O during cooling in an ice-bath. Filter it off and dry it at 105o. [Beilstein 13 IV 2283.]
