Add 581g of tert-butyl acetate and 147g of glutamic acid to a 2000mL three-necked flask, stir, add 100mL of perchloric acid dropwise, react at 20℃ for 48 hours, cool to 0℃, add 600mL of water, neutralize with Na2CO3 to pH=8-9, separate the phases, wash three times with 100mL of 1% Na2CO3 aqueous solution, combine the aqueous phases, and extract three times with 200mL of petroleum ether to obtain crude product containing L-Glutamic acid α-tert-butyl ester.

L-Glutamic acid α-tert-butyl ester (CAS 45120-30-7) is a high-purity, asymmetrically protected amino acid building block. Its synonyms include: H-Glu-OtBu, L-Glu-OtBu, Glu-OtBu, 1-tert-butyl L-glutamate and glutamic acid tert-butyl ester, amongst others. The α-position of this molecule contains a carboxyl group protected by a tert-butyl ester, whilst the carboxyl group at the other end is free and can be used to attach side chains for the preparation of long-acting GLP-1 analogues or for PROTAC synthesis.
