What is the HBTU coupling mechanism?
Aug 18,2025
HBTU is a widely used peptide coupling reagent for both solid-phase and solution reactions and is the preferred reagent for coupling phosphorylated amino acids. Its coupling mechanism is via activation of the carboxyl group (-COOH) to form a reactive intermediate, an activated ester, which is then attacked by an amine to form the desired peptide bond.
