Bioactivity

名称	5-BrdU
描述	5-BrdU (Broxuridine) is a nucleoside analogue. 5-BrdU is used to detect proliferating cells and compete with thymidine for incorporation into DNA.
细胞实验	Cultures are initially plated at 2000 cells/cm2 and are quantified with a Z2 Coulter Counter. RG2 rat glioma cells are treated once with 0, 1, 10, or 50 μM BrdU for 24 hours, and cumulative growth curves were obtained over 18 days. Control and treated cells are quantified and replated at equal densities on days 5, 12, and 18 after treatment.(Only for Reference)
激酶实验	Assays are run in the presence of 100 μM ATP using 10 μM of substrate. 30 μL PROTEIN-MIX in 25% DMSO and incubated for 15 min at room temperature. 10 μL PEPTIDE-MIX is added, the mixture is incubated for 60 min at RT and stopped by adding 180 μL 6.4% TCA (final concentration: 5%). Incorporated phosphate is measured in a scintillation counter and IC50 values are calculated using a sigmoidal curve analysis program with variable hill slope[1].
体外活性	方法：小鼠神经前体细胞用5-BrdU处理，使用MTT检测细胞增殖活性。结果：5-BrdU的抗增殖活性EC50值为2.045μM。[1] 方法：L5178Y小鼠白血病细胞用5-BrdU处理，通过生长抑制测定细胞生长抑制情况。结果：在L5178Y小鼠白血病细胞中，5-BrdU的IC50值分别为20μM（通过[14C]亮氨酸掺入测定）和26μM（通过[14C]胸苷掺入测定）。[2]
体内活性	方法：为研究5-BrdU的抗肿瘤活性，在大鼠胶质瘤RG2肿瘤模型中将5-BrdU腹腔注射（300 mg/kg）或口服给药（0.8 mg/mL）。结果：5-BrdU显著减缓了肿瘤的进展。[3] 方法：为研究胚胎期暴露对小鼠小脑神经上皮的影响，怀孕的大鼠在胚胎第13天时，单次注射不同剂量（25-300 µg/g）的5-BrdU。结果：低剂量（25-75 µg/g）的5-BrdU并未改变小脑神经上皮细胞的增殖行为，而高剂量（100-300 µg/g）则导致相关增殖参数降低，其中300 µg/g剂量的影响最为显著。[4] 方法：为研究5-BrdU对小鼠小脑外颗粒层神经母细胞的影响，在小鼠出生后第9天，单次注射不同剂量（50-300 µg/g）的5-BrdU。结果：不同剂量的5-BrdU会影响细胞周期各阶段的持续时间及整个细胞周期的完成时间。[4] 方法：为研究5-BrdU对成年大鼠海马齿状回神经发生的影响，成年大鼠接受累积剂量的5-BrdU（40、240或460 mg/kg）处理。结果：5-BrdU并未减少海马齿状回的细胞增殖或未成熟神经元的数量。[5]
