Bioactivity

名称	Myricetin
描述	Myricetin (Cannabiscetin) is a natural flavonoid and MEK1 inhibitor. Myricetin has hypoglycemic, antioxidant, hepatoprotective, anti-tumor and anti-inflammatory activities.
细胞实验	Pancreatic cancer cells (MIA PaCa-2, Panc-1 or S2-013) or normal pancreatic ductal cells (PDCs) are treated with myricetin (12.5–200 μM). Cell viability is determined using the Dojindo Cell Counting Kit-8. Cells are seeded onto a 96-well plate at 1×104 cells per well and allowed to adhere overnight. After treatment with myricetin at various concentrations for 24 hours, 10 μL of the tetrazolium substrate is added to each well of the plate. Plates are incubated at 37°C for 1 hour, after which the absorbance at 450 nm is measured[2].
体外活性	方法: 人肝癌细胞 HepG2 和 Huh-7 用 Myricetin (100-200 µM) 处理 72 h，使用 CCK-8 assay 检测细胞活力。结果: Myricetin 以剂量依赖性方式显着抑制细胞生长。[1] 方法: 卵巢癌细胞 A2780 和 OVCAR3 用 Myricetin (25 µM) 处理 48 h，使用 single stranded-DNA Apoptosis ELISA kit 检测细胞凋亡。结果: 在用 Myricetin 处理的 A2780 细胞中，与未处理的细胞相比，观察到凋亡信号增加了约 2.5 倍，而在类似的处理条件下，OVCAR3 细胞中的凋亡信号与未处理细胞相比增加了约 4 倍。[2]
体内活性	方法: 为检测体内抗肿瘤活性，将 Myricetin (30 mg/kg，每天一次) 和 cisplatin (5 mg/kg，每三天一次) 腹腔注射给携带 Huh-7 异种移植物的 BALB/c 裸鼠，持续两周。结果: 单独用 Myricetin 或 cisplatin 治疗适度抑制肿瘤生长，但联合治疗比单独用 Myricetin 或 cisplatin 更明显地抑制肿瘤生长。[1]

Dissolve by heating and/or sonication if necessary. Working solution is recommended to be prepared and used immediately.
