名称	Meropenem
描述	Meropenem (SM 7338) is a synthetic carbapenem antibiotic with broad-spectrum antibacterial activity. It exerts a bactericidal effect by inhibiting the synthesis of peptidoglycan in the bacterial cell wall and is active against a wide range of Gram-positive, Gram-negative, and anaerobic bacteria. Meropenem is active against both susceptible and resistant Neisseria gonorrhoeae (MIC values of 0.02–0.06 mg/mL), Haemophilus influenzae (MIC: 0.03–0.12 mg/mL), and Klebsiella pneumoniae (MIC: 0.015–0.12 mg/mL). Meropenem can be used for constructing bacterial infection models and screening for antimicrobial activity.
激酶实验	Complementation of SCR7 Inhibition with Puri?ed Ligase IV: Complementation experiment is carried out by adding increasing concentrations of puri?ed Ligase IV/XRCC4 complex (30, 60, and 120 fmol) along with the oligomeric DNA substrates (5' compatible and 5'-5' noncompatible ends) to the SCR7-treatedextracts. Reactions are incubated for 2 h at 25℃. The reaction products are then resolved on 8% denaturing PAGE. The gel is dried and exposed and the signal is detected with a PhosphorImager and analyzed with Multi Gauge (V3.0) software.
体外活性	方法：产 NDM-1 的大肠杆菌 ZC-YN3 临床分离株以及产 NDM-5、NDM-9 的菌株加入 Meropenem 单药或厚朴酚单药或 Meropenem + 厚朴酚（32 μg/mL）联用，检测最低抑菌浓度（MIC）。结果： Meropenem 单药 MIC：16 μg/mL （对 ZC-YN3），厚朴酚单药 MIC：>1024 μg/mL （无直接抗菌活性），Meropenem+厚朴酚联用 MIC：4 μg/mL （对 ZC-YN3），降低 4 倍。[1] 方法：人类原代肺泡上皮细胞（HPAEpiC），经 FlexCell 系统模拟机械拉伸后感染 PAO1 菌株。在感染后 2 小时加入药物加入 Meropenem （5 × MIC，0.5 × MIC），在感染后 4 小时和 8 小时评估细菌载量。结果：Meropenem 有效减少细菌载量。[2]
体内活性	方法：C57BL/6J 小鼠，机械通气后经气管感染 PAO1 菌株，感染 4 小时和 16 小时后各给药一次，腹腔注射 Meropenem 单药。结果：Meropenem 单药能显著降低肺部及全身细菌载量[2]

溶解度	10% DMSO+40% PEG300+5% Tween 80+45% Saline : 4 mg/mL (10.43 mM), Sonication is recommended. DMSO : 100 mg/mL (260.78 mM), Sonication is recommended. Ethanol : < 1 mg/mL (insoluble or slightly soluble) H2O : 5 mg/mL (13.04 mM), Sonication is recommended.
