专业生产工艺链(Staherb SOP 概要)

  1. 前体准备(双轨)


    • 外采路线:定点基地柚/葡萄柚/枳实幼果皮渣 → 60%–80% 乙醇提 → AB-8 大孔富集 → 水/乙醇重结晶 → Naringin 98% HPLC 作投料(Staherb 双轨溯源);


    • 入厂拒收:Naringin 初含量 <3% DW、硫熏 SO₂>150 ppm、农残超 GB 2763、霉变皮。


  2. 碱液开环(查尔酮中间体):Naringin 投料溶于 4%–15% KOH 或 NaOH 水溶液(柚皮苷:碱质量比 ~3:1.5,终 pH~13),40–80℃ 搅溶 30–70 min,TLC/UV 监黄→查尔酮位移(C2=C3 开环),滤液除不溶物;可补 5%–50% 乙醇/甲醇助溶(文献 CN105801636A 参 )。


  3. 催化加氢(C2=C3 饱和):开环液进高压釜,加 10% Pd/C(投料 1%–20% w/w 柚皮苷,主用 5%–10%) 或 Raney Ni,通 H₂ 0.5–3 MPa(主用 0.5–1.2 MPa)、20–60℃(主用 50℃)加氢 5–12 h(主用 6–8 h),至不吸氢;滤除 Pd/C(再生/危废合规处置),氢化液无双键(HPLC 单峰)。


  4. 酸化析晶与重结晶(A 线):氢化液 HCl/H₃PO₄ 调 pH 5–7,搅拌降温 <20℃ 析晶,离心得 NDHC 粗品;粗品以 15% 乙醇 6 倍量热溶 + 1%–4% 活性炭脱色 25 min → 冷却 4–20℃ 重结晶 2–3 次 → 60℃ 真空干燥(避光 N₂)98%/99% HPLC 白晶;低标 90%/95% 可跳最后一次重结晶直接干法标化。


  5. 物理成型(B 线,选做):98%+ 晶体 → 气流微粉(D90≤10 μm 速溶)或预溶入热水/丙二醇得定制浓度水溶液(食品客户后段直添);与赤藓糖醇/甜菊糖苷预混矫苦包(微胶囊化)。


  6. 筛分与质控:80–100 目旋振筛 → 同标多批 V 混 → HPLC-UV@282 nm 外标报纯度% + 特征杂质 Naringin/开环查尔酮/柚皮素残留检 + 甜度比对(5% 蔗糖参比 300–500×)+ 亚阈掩苦模型(柚皮苷苦液)+ Pd≤5 ppm/重金属 Pb≤1 Cd≤0.3 As≤1/Hg≤0.1 + EtOH≤500 ppm + 微生物 TPC≤1000 cfu/g → 金属探测。

Sensitive redlines: hydrogenation involves H₂ high-pressure (0.5–3 MPa) + Pd/C noble-metal catalyst, restricted process; finished must control Pd≤5 ppm, EtOH≤500 ppm, no NaOH/KOH residue (pH 5–7 wash-crystallized); NDHC slightly hygroscopic, free-form sealed 2–8℃ dry dark 24 mo.

Dedicated Production Chain (Staherb SOP summary)

  1. Precursor prep (dual-track): external route — registered-base pomelo/grapefruit/zhishi peel-dreg → 60%–80% EtOH extract → AB-8 enrich → H₂O/EtOH recrystallize → Naringin 98% HPLC feed (Staherb dual genealogy); reject Naringin initial <3% DW, SO₂>150 ppm, pesticide over GB 2763, mold.


  2. Alkaline ring-open (chalcone intermediate): Naringin in 4%–15% KOH or NaOH aq (naringin:alkali ~3:1.5 w/w, final pH~13), 40–80℃ dissolve 30–70 min, TLC/UV monitor yellow→chalcone shift (C2=C3 open), filter insolubles; optional 5%–50% EtOH/MeOH cosolvent (ref CN105801636A ).


  3. Catalytic hydrogenation (C2=C3 saturate): ring-open liquor to autoclave, add 10% Pd/C (1%–20% w/w naringin, mainly 5%–10%) or Raney Ni, H₂ 0.5–3 MPa (mainly 0.5–1.2 MPa), 20–60℃ (mainly 50℃), 5–12 h (mainly 6–8 h) till no H₂ uptake; filter Pd/C (regenerate/compliant hazardous waste), hydrogenate liquor single HPLC peak.


  4. Acid crystallize & recrystallize (Line A): hydrogenate liquor HCl/H₃PO₄ to pH 5–7, stir cool <20℃ crystallize, centrifuge NDHC crude; crude in 15% EtOH 6× hot-dissolve + 1%–4% activated carbon 25 min → cool 4–20℃ recrystallize 2–3× → 60℃ vacuum dry (dark N₂) to 98%/99% HPLC white crystal; 90%/95% can skip last recrystallization direct dry-standardize.


  5. Physical forming (Line B, optional): 98%+ crystal → jet micronize (D90≤10 μm instant) or pre-dissolve in hot-water/propylene glycol to custom conc (beverage post-stage); pre-blend with erythritol/stevia bitter-cut microcap.


  6.
