氮杂环卡宾(N-heterocyclic carbene,NHC)钯催化剂是一类以氮杂环卡宾为配体、钯为金属中心的过渡金属催化剂,凭借配体与金属中心的强配位作用、优异的稳定性及可调的催化活性,在有机合成领域占据重要地位,尤其在碳-碳、碳-杂原子键构建反应中展现出独特优势。该催化剂在交叉偶联、C-H键活化、羰基化、脱氟烷基化等反应中均有不错的应用。以下为其部分应用实例:
以SIPr-PdCl2-Py为催化剂,末端炔烃与芳基三甲基铵三氟甲磺酸盐可通过Sonogashira 交叉偶联反应,一锅法直接实现胺的炔基化,无需分离铵盐[1]。

以[Pd(NHC)(acac)Cl]为催化剂,可通过选择性N−C/O−C键断裂,高效催化酰胺和酯类的Suzuki−Miyaura及Buchwald−Hartwig交叉偶联反应[2]。

在Pd(OAc)2/S-IPr催化下,酮类化合物可与(E)-偕烷基、芳基二取代烯丙基碳酸酯发生烯丙基烷基化反应,构建含邻位叔碳和季碳立体中心的酮类产物[3]。

以Pd-PEPPSI-IPentAn配合物为催化剂,可在有氧条件下实现(杂)芳基氯与胺的C−N 交叉偶联反应,高效构建CAr−N键[4]。

以Pd(IPr)(cinammyl)Cl为催化剂,芳基酯可发生C−O键断裂,与硼酸进行Suzuki−Miyaura 偶联反应制备含羰基的酮类产物[5]。

在Pd-PEPPSI-IPr催化下,芳基酰胺进行C-N键活化,与芳基硼酸发生Suzuki−Miyaura 交叉偶联反应制备酮类化合物[6]。

以Pd/IPrBIDEA作为催化剂,以3,3-二甲基烯丙基频哪醇硼酸酯作为新型氢化物供体,偕二氟环丙烷可发生开环氢脱氟反应,高效高区域选择性合成热力学上较不稳定的末端氟代烯烃[7]。
