Introduction：
Diphenylphosphine oxide is an important intermediate in organic synthesis, widely used in the synthesis of various pesticides and chiral phosphine ligands, and can replace alkali metal cyanide as a coupling agent for the synthesis of heterocyclic compounds, such as the synthesis of herbicide paraquat under mild conditions.
Application:
Diphenylphosphine oxide is commonly used in the preparation of triphenylphosphine oxide, alkenyl diphenylphosphine oxide, addition to alkynes, and Wittig-Horner reaction reagents. The synthetic Ph2P(O)H of diphenylphosphine derivatives is coupled with aryl trifluoromesylate, and the reduction of phosphine oxides to diphenylaryl phosphine is a common chiral ligand.
Synthesis methods:
There are three main synthesis methods of diphenyl phosphine oxide. Method 1 is based on POCl3 as raw material, which is reduced to lithium aluminum hydride by Fuchs alkylation reaction. Method 2 uses diethyl phosphite as raw material and reacts with Grignard reagent to produce the product; In Method 3, Miller et al. took PCl3 as raw material and obtained the target product by proper hydrolysis after Fuke alkylation reaction. However, the purification of the intermediate Ph2PCl was difficult and the yield was only 37.0%.
