四丁酸核糖黄素酯 用途与合成方法
取试样10rag溶于5ml乙醇,加由15%盐酸羟胺溶液和15%氢氧化钠溶液的1:1混合液2ml,充分摇匀,再加O.8ml盐酸和O.5ml氯化铁试液(TS-101)及等体积的乙醇后,应呈深红棕色。 将试样在真空干燥器(硫酸)中干燥4h后,精确称取约40mg,溶于乙醇并稀释至500mI,取10ml,加乙醇至50ml,作为试样液。将其盛于1cm比色池中,测定在波长445nm处的吸光度A1,以乙醇为对照液。另取标准核黄素在105℃下干燥3h后,精确称取约50mg,加冰醋酸4ml和水150ml的混合液,加热溶解,冷却后加水至500ml。取该液5ml,加乙醇至50ml,作为标准液。与试样液一样,测定标准液在波长445nm处的吸光度A2,按下式求取核黄素四丁酸酯(C33H44O10N4)的含量。
含量(%)=(A1/A2)×[25X1.745/式样量(mg)]×100 毒性极低。LD50>15g/kg(小鼠,经口)。大鼠按500~2000mg/kg授予6个月,无明显变化。 GB 14880—94:同“01022,核黄素”。 Riboflavin tetrabutyrate是亲脂性黄素衍生物,具有抗氧化和移除脂类过氧化物活性。
Riboflavin Tetrabutyrate inhibits oxygen uptake by lipid peroxidation. Riboflavin Tetrabutyrate is suppressive against both NADPH-coupled and ascorbate-induced microsomal lipid peroxidation. Riboflavin Tetrabutyrate seems to exhibit its antioxidative action at or after the hydrogen atom is abstracted as a free radical from an active methylene group of polyunsaturated fatty acids during the process of enzymic oxidation-reduction reaction.
Riboflavin Tetrabutyrate might improve the metabolism of lipids in patients suffering from atherosclerosis, diabetes, fatty liver and so on through the inhibition of lipid peroxide, resulting in the decrease of the elevated serum lipid. Feeding of Riboflavin Tetrabutyrate results in an increase in the hepatic activity of 3-ketoacyl-CoA thiolase by 50% of the control level, while the activities of renal 3-ketoacyl-CoA thiolase and of hepatic and renal acyl-CoA synthetase and acyl-CoA dehydrogenase remain unaffected. The increase in hepatic 3-ketoacyl-CoA thiolase activity suggests that prolonged Riboflavin Tetrabutyrate administration results in an increased beta-oxidation of fatty acid in the liver .
