以5-羟基烟酸甲酯为原料合成6-氯-5-羟基烟酸甲酯的一般步骤如下:在冰浴冷却条件下,向含有5-羟基烟酸甲酯(4.5g,29mmol)的烧瓶中缓慢滴加次氯酸钠水溶液(6.15%,26.7mL,22mmol)。反应混合物搅拌30分钟后,加入2M盐酸(20mL)以终止反应。通过过滤收集反应中生成的白色晶体,得到6-氯-5-羟基烟酸甲酯2.31g,收率为42%。产物结构通过1H NMR(400MHz,CDCl3)和LC-MS进行确认:1H NMR δ10.32(bs,1H),8.48(d,1H),7.84(d,1H),7.40(s,1H),3.93(s,3H);LC-MS(ELSD)显示C7H6ClNO3 [M]+的计算质量为187,实测值为187。
