Step-2: Potassium tert-butoxide (23.4 g, 299.4 mmol) was dissolved in 200 mL of tert-amyl alcohol under nitrogen protection and stirred at room temperature. Subsequently, a solution of 2-chloro-N-(2,3-dihydroxypropyl)acetamide (Step-1 product of Intermediate-9) (20 g, 119.6 mmol) dissolved in 460 mL of tert-amyl alcohol was added to this solution and the reaction lasted for 2 hours. After 1 hour of reaction, methanol (100 mL) and water (3 mL) were added and stirring was continued for 20 minutes. Upon completion of the reaction, the reaction mixture was concentrated under vacuum and purified by rapid chromatography on silica gel using methanol/ethyl acetate (20:80) as eluent to afford 6-(hydroxymethyl)morpholin-3-one as a white solid (14.4 g). Mass spectral data: m/z 132.1.
