N-substituted morpholines contain dual groups of tertiary amine and ether, making them important catalysts not only in the semi-synthetic production of penicillin and polyurethane foaming, but also crucial raw materials for the synthesis of N-substituted oxidized morpholines. Current synthetic processes mainly include the morpholine method, the N-methyldiethanolamine method, the diethanolamine method, the diethylene glycol method, and the dichloroethyl ether method. The substitution of 3(S)-HYDROXYMETHYL-4-BOCMORPHOLINE is prepared by reacting (R)-(4-benzyl-3-morpholino)-methanol with ditert-butyl dicarbonate. The synthetic reaction formula is shown in the figure below:

Experimental procedure:
Triethylamine was added to a dichloromethane solution of (R)-(4-benzyl-3-morpholino)-methanol, followed by dropwise addition of a dichloromethane solution of ditert-butyl dicarbonate. After the addition was complete, the reaction was allowed to proceed for 15 hours. The solvent was removed by vacuum distillation, and the concentrate was purified by silica gel column chromatography [eluent: V (ethyl acetate) / V (petroleum ether) = 1/10] to obtain the compound 3(S)-HYDROXYMETHYL-4-BOCMORPHOLINE.
