2-Chloro-4-trifluoromethylpyrimidine (2.0 g, 11 mmol) and piperazine (2.8 g, 33 mmol) were dissolved in DMF (10 mL) and triethylamine (3.0 mL, 22 mmol) was added as base. The reaction mixture was placed in a sealed tube and the reaction was stirred at 100 °C overnight. After completion of the reaction, most of the solvent was removed under reduced pressure. The crude product was purified by silica gel column chromatography with EtOAc to EtOAc/MeOH/NEt3 (9:1:0.5) as eluent to afford 1-(4-trifluoromethylpyrimidin-2-yl)piperazine (1.48 g, 56% yield). Mass spectrometry analysis: the calculated value (M+H) of C9H11F3N4 was 233 and the measured value was 233.1.
