Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HeLa (Human epithelial cells from cervix adenocarcinoma) cells labeling JNK1+JNK2+JNK3 with ab179461 at 1/250 dilution, followed by Goat anti-rabbit IgG (Alexa Fluor® 488) (ab150077) secondary antibody at 1/400 dilution (green). Confocal image showing both cytoplasmic and nuclear staining on HeLa cells. The nuclear counterstain is DAPI (blue). Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution and ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution (red).
The negative controls are as follows:- -ve control 1: - ab179461 at 1/250 dilution followed by ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution. -ve control 2: - ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution followed by ab150077 (Alexa Fluor®488 Goat Anti-Rabbit IgG H&L) at 1/400 dilution.
C-Jun N末端激酶(JNK)也称为丝裂原激活的蛋白激酶(MAPK8)。它属于应激激活蛋白激酶的MAPK超家族。MAPK是丝氨酸-苏氨酸蛋白激酶,可响应多种细胞外刺激而被激活,并介导从细胞表面到细胞核的信号转导。JNK参与多种细胞过程,例如增殖,分化,转录调节和发育。JNK途径受压力和炎症信号的刺激。由于差异的mRNA剪接,JNK被表达为十种不同的亚型。主要形式为JNK1和JNK2。 JNK123能够使c-Jun(一种转录调节因子,属亮氨酸拉链家族成员)氨基末端活性区Ser63和Ser73发生磷酸化的蛋白激酶。是促分裂原活化的蛋白激酶超家族中的一个家族,包括JNK1、JNK2、JNK3,当其活化模体Thr-Pro-Tyr内的Thr、Tyr被JNKK1、JNKK2磷酸化后而激活,活化酶可磷酸化c-Jun等多种蛋白质,从而影响细胞凋亡、炎症和肿瘤的发生。 JNK123抗体是一类可以特异性结合JNK123的多克隆抗体,主要用于检测JNK123的Western Blot、IHC-P、IF、ELISA、Co-IP等多种免疫学实验。 双抗体夹心法测定标本中JNK123水平。用纯化的JNK123抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入JNK123,再与HRP标记的JNK123抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的JNK123呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),通过标准曲线计算样品中JNK123浓度。
