Chemical Properties
This product is a yellow to orange-red clarified liquid; slightly peculiar fishy odor, but no oil odor. It can be mixed with ether and chloroform arbitrarily, slightly soluble in ethanol. Sodium cod liver oil injection purified by saponification with cod liver oil as raw material is a kind of preparation used for the treatment of internal hemorrhoids, hemangioma and varicose veins of the lower limbs.
Definition
An oil containing a high percentage of vitamin A. High-potency livers, as from cod, shark, and halibut, contain from 100,000 to 1,500,000 A units/g. The oil is extracted by cooking the livers under low-pressure steam and removing the oil, which floats on
Synthesis
1) Extraction: weigh 200kg of grinded tuna liver, add 400kg of ethyl acetate, stirring, extraction for 80 minutes, take the ethyl acetate layer; again with 240kg of ethyl acetate extraction for 80 minutes, take the ethyl acetate layer; combined, the ethyl acetate extraction phase 625L;
2) Salt precipitation: add 125L saturated Na2CO3 in the ethyl acetate extraction phase, adjust PH to 8, stir for 5 minutes, leave for 0.5h;
3)Acid washing: after washing three times with 0.1NHCL, add Na2SO4 according to 1kgNa2SO4/50L ethyl acetate and leave overnight;
4)Distillation: under the condition of 40-50, distillation and concentration for 10h, get 40L of fish oil; under the condition of 85, rotary evaporation and distillation for 12h, get 5.3L of crude fish oil;
5) acetone refining: in the fish oil crude add 10L acetone, mix well, under the condition of 0-5 , after standing for 2.5h, filtration, filtrate distillation and concentration, to get 4.7L of fish oil;
6) molecular distillation: the fish oil obtained in step 5) is subjected to two-stage molecular distillation, in which the first stage of molecular distillation, the low temperature is 80 , the high temperature is 170 , distillation 8h; the second stage of molecular distillation, the low temperature is 80 , the high temperature is 195 , distillation 9h, to obtain 4.3L of fish oil;
7) column chromatography: after dissolving the fish oil obtained in step 6) in 4.3L of ethyl acetate, after glass chromatography column treatment, take the ethyl acetate chromatogram, distillation and concentration, to obtain 4L of cod liver oil.
8) Freezing separation: the fish oil produced in step 7) was placed in the freezing at 5 for 5 hours, and at 5 , press filtration by plate and frame filter press to produce 3.9L of cod liver oil.
