N-Methylisatoic anhydride is a 2'-OH selective acylation agent of RNAs, and is widely used for resolving secondary RNA structures using the SHAPE (Selective 2'-Hydroxyl Acylation Analyzed by Primer Extension) technology.

Application
N-Methylisatoic anhydride was first condensed with ethanolamine to give N-(2-hydroxyethyl)-2-methylaminobenzamide. Cyclization of the latter with ethyl pyruvate gave quinazoline derivatives which, upon hydrolysis and dehydrative cyclization with l-methyl-2-chloropyridinium iodide, afforded the l,4-oxazines[3,4-]quinazoline.
