具体操作方法如下:使用3L三颈烧瓶进行反应。将(S)-4-碘-L-苯丙氨酸(200.00g,687.10mmol,1.00当量)、1,4-二恶烷(1.00L)和水(1.00L)依次加入反应瓶中,随后加入氢氧化钠(68.71g,1.72mol,2.50当量),溶液逐渐澄清,温度轻微上升至19℃。待体系冷却至0-10℃后,缓慢滴加二碳酸二叔丁酯(254.93g,1.17mol,268.35mL,1.70当量),反应温度自然升至10-30℃。加毕,反应体系于室温(约30℃)下搅拌8小时。反应进程通过高效液相色谱(HPLC)监测,直至原料完全转化。随后,控制体系温度低于40℃,减压浓缩除去1,4-二恶烷。待反应混合物冷却至室温(约25℃)后,加入100mL水淬灭反应,并用2M盐酸调节pH至1.8-2.0。所得混合物用乙酸乙酯(2L×3)萃取三次。合并有机相,依次用饱和盐水(1L×2)洗涤两次,无水硫酸钠(200g)干燥,减压浓缩得淡黄色固体粗产物。粗产物于40-45℃烘箱中干燥,得到白色固体(S)-N-Boc-4-碘-L-苯丙氨酸(250.00g,626.28mmol,产率93.00%,HPLC纯度98%)。产物结构经1H NMR(400MHz, DMSO-d6)确证:δ 7.49(d, J = 7.8Hz, 2H),6.88(d, J = 7.8Hz, 2H),5.80(d, J = 5.9Hz, 1H),3.68(d, J = 5.5Hz, 1H),3.00-2.90(m, 1H),2.87-2.75(m, 1H),1.35-1.15(m, 9H)。
