以甲磺酸酯17(1.2g,3.78mmol)为原料,将其悬浮于无水THF(38mL)中,加入三苯基膦(1.48g,5.66mmol),室温搅拌反应2小时。随后加入H2O(0.5mL),继续搅拌18小时。反应完成后,减压除去溶剂,将残余物悬浮于1M盐酸(150mL)中,用CH2Cl2(2×70mL)萃取。水层用碱调节至pH 14后,再用CH2Cl2(3×70mL)萃取。合并有机相,用盐水(100mL)洗涤,无水MgSO4干燥,减压浓缩。通过硅胶快速色谱法纯化,洗脱剂为CH2Cl2-甲醇-氨水(90:9:1),得到目标产物胺18(0.52g,2.62mmol,收率71%)为无色固体。产物表征数据如下:熔点76.8-77.9℃;比旋光度[α]D -47.6(c 1.0,CHCl3);红外光谱(νmax/cm-1)2975、1688、1408、1170;1H NMR(500MHz,CDCl3)δ 4.43(0.5H,s)、4.30(0.5H,s)、3.69(1H,s)、3.38-3.32(1H,m)、3.19-3.13(1H,m)、3.06-2.97(2H,m)、1.75-1.65(2H,m)、1.45-1.43(9H,m);13C NMR(125MHz,CDCl3)δ 154.6、154.3、79.6、79.5、57.3、56.6、56.5、56.3、56.1、52.8、52.7、37.2、37.0、28.66;质谱(APCI)m/z 199([M+H]+,100)。
