2,5-Diaminoanisole sulfate is cyclized with glycerol in the presence of iodine and potassium iodide to generate 6-methoxy-4,7-phenanthroline. Further oxidation can obtain 4,7-phenanthroline-5,6-diquinone, which is an anti-amebic dysentery and bacillary dysentery drug, and can also be used in hair dyes and organic pigment intermediates.

The o-acetamido methyl ether is nitrated with nitric acid and reduced with iron powder to obtain 5-amino-2-acetamidoanisole, and then heated to reflux with sulfuric acid at 100-104°C for 0.5h, and hydrolyzed to obtain 2,5-Diaminoanisole sulfate.
