SW063058 is an autophagy inducer that specifically disrupts the binding of Beclin 1 to Bcl-2 while leaving the interaction between Bcl-2 and pro-apoptotic members like Bax and BIM unaffected. By inhibiting the negative regulatory effects of Bcl-2 on Beclin 1, which is crucial for the initiation of autophagy, SW063058 promotes autophagic activity without triggering cytotoxicity, apoptosis, or other forms of cell death in vitro.
Biological Activity
SW063058 is an autophagy inducer th at selectively disrupts Beclin 1/Bcl-2 bindingversus Bax/Bcl-2 binding. SW063058 acts by disrupting the binding of Bcl-2a negative regulator of autophagyto the BH3 domain of Beclin 1which is involved in autophagy initiation. It does not inhibit Bcl-2 binding to the BH3 domain of the pro-apoptotic Bcl-2 family members BAX and BIM. In vitro experiments showed th at SW063058 was not cytotoxic and induced autophagy without inducing apoptosis or other forms of cell death.
