6-Fluoro-2-methylindan-1-one is a key intermediate in the synthesis of sulindac. This compound is converted to sulindac via hydrolysis following condensation with cyanoacetic acid, followed by condensation with p-methylthiobenzaldehyde and subsequent oxidation. Sulindac is a non-steroidal prodrug with minimal biological activity; once it enters the body, it is metabolised into an active sulphide compound. This compound inhibits cyclooxygenase and reduces the synthesis of prostaglandins, thereby exerting analgesic, anti-inflammatory and antipyretic effects. It is primarily used to treat pain, rheumatoid arthritis, ankylosing spondylitis and gouty arthritis.
