Biosensor detects NADH trimetabolite for disease monitoring

Description of NADH, disodium salt

NADH disodium salt is a donor of ADP-ribose units in the ADP-ribosylaton reaction and a precursor of cyclic ADP-ribose. NADH disodium salt acts as a regenerative electron donor in cellular energy metabolism processes such as glycolysis, β-oxidation and the tricarboxylic acid (TCA) cycle. As an efficient electron proton donor, reduced nicotinamide adenine dinucleotide (NADH) coenzyme holds great promise in enzymatic industry.

Uses of NADH

Nicotinamide adenine dinucleotide (NADH) plays a pivotal role in metabolism. Convenient detection NADH its related metabolites has pursuit point-of-care clinical analysis. Here, we propose a polymer dots (Pdots)-based NADH-sensitive electrochemiluminescence (ECL) biosensor for detection NADH three metabolites. Pdots acted as efficient ECL emitters without additional modification to construct this biosensor. Specially, NADH both acted as final detection target at same time as bio-coreactants to sensitively influence ECL intensities, in which NADH was generated or consumed in presence target analyte their specific enzyme. For glucose lactic acid detection, NAD+ was reduced to NADH to generate an enhanced ECL signal. Conversely, for pyruvate detection, NADH was consumed to further decrease ECL. The designed Pdots-based ECL biosensor showed wide detection ranges, high selectivity low limits detection 4.6 μM, 0.7 μM 0.5 μM for analysis three analytes, respectively. This strategy was successfully applied in quantifying concentrations glucose, lactic acid pyruvate in human serum, which also has potential to be implemented as a powerful fast tool for ECL sensing NADH other related metabolites for point-of-care use disease monitoring.
