Cardamonin is a chalcone occurring in many plants and is reported to have a marked anti-tumor activity. Some reports indicates that cardamonin has inhibitory effect on NO generation and is associated with the inhibition of IκB kinase (IKK)in LPS-stimulated RAW264.7 Mφ.
In the newest paper in Life Science, Takahashi et al. explored the possible relationship between cardamonin and NOS/NO signaling pathway. Treatment of cardamonin and related compounds, flavokawain B and naringenin chalcone, all caused marked NO suppression in RAW264.7 mouseMφ. On the other hand, in vivo assay also demonstrated that cardamonin inhibit LPS-induced NO production. The molecular mechanisms was further studied and the result showed that pre-treatment with cardamonin blocked STAT-1 activation and thus inhibit the expression of iNOS mRNA expression. Moreover, cardamonin was found to suppress IFN-γ production. Since IFN-γ was demonstrated to mediate LPS-induced STAT-1 phosphorylation and NO production , this may be the reason of decreasing NO generation[1].
In summary, cardamonin can suppress LPS-induced NO generation by downregulated IFN-γand block STAT pathway. Therefore, Cardamonin may be a potential therapy against inflammatory and autoimmune diseases.
