GW501516 also known as GW-501,516, GW1516 and GSK-516.It is a selective agonist (activator) of the PPARδ receptor. It was entered into clinical development as a drug candidate for metabolic diseases and cardiovascular diseases.

GW501516 is a synthetic ligand of PPARδ and belongs to the phenoxyacetic acid derivative. After binding to GW501516, activated PPARδ forms a heterodimer with 9-cis retinoid receptor (RXR) And then identify the peroxisome proliferator response element (PPRE) upstream of the target gene promoter to regulate gene transcription, translation and biological effects downstream of the target gene.

GW501516 inhibits apoptosis of ox-LDL or high glucose-induced HUVECs and attenuates the inhibitory effect of ox-LDL or high glucose on HU-VECs activity.
