名称EGFR-IN-47
描述EGFR-IN-47 (compound 14aj) is a potent, fourth-generation, and orally active EGFR inhibitor. It specifically targets the EGFR L858R/T790M/C797S triple mutation with an IC50 of 0.01 uM. In NSCLC research, EGFR-IN-47 effectively overcomes C797S-mediated resistance to Osimertinib by inhibiting the phosphorylation of EGFR and its downstream mediators, subsequently inducing cell cycle arrest and apoptosis.
体外活性EGFR-IN-47 强效抑制 PC-9 (L858R/T790M/C797S) 细胞生长 (IC50 = 0.013 uM)，可诱导 G0/G1 期停滞 [1]。
体内活性EGFR-IN-47 (10-40 mg/kg p.o.) 在异种移植瘤模型中表现出显著活性；大鼠 PK 研究显示其生物利用度为 66.05% [1]。
