Bioactivity

名称Neutral protease, Paenibacillus polymyxa
描述Neutral protease, Paenibacillus polymyxa is a neutral protease with strong fibronectinase and type IV collagenase activities. It can be used to separate intact epidermis from dermis and to detach intact epithelial sheets cultured in vitro from their substrates.
体外活性使用方法：1. 溶液配制：使用DPBS缓冲盐溶液（无钙镁离子）将冻干粉溶解制备10 mg/mL的储存液，并使用0.22 μM滤膜进行除菌过滤。使用时，用DPBS稀释储存液至工作浓度，细胞分离常用工作浓度为0.6-2.4 U/mL。注意：工作浓度不宜超过2.4 U/mL。2. 组织解离：利用无菌刀或剪刀将组织剪成3-4 mm的块状，使用无菌PBS清洗，然后加入Dispase II溶液（0.6-2.4 U/mL）确保完全浸没，并在37℃下孵育，孵育期间轻微搅拌，直到组织解离。对于难以解离的组织，一般1小时即可完全分离，但长时间（如数小时）孵育对细胞活性无明显影响。有需要时，可用无菌不锈钢网筛过滤消化产物，以分离单细胞和残留组织块；或者在大块组织沉淀后轻轻倒出上层细胞，必要时可使用新鲜dispase进一步解离。离心收集细胞并弃去酶溶液，用培养基重悬细胞沉淀，并在常规条件下培养。3. 细胞传代：用37℃预热的Dispase溶液处理细胞，于37℃孵育5分钟后吸除溶液，继续孵育10分钟，观察显微镜下的细胞分离情况，必要时可延长孵育15分钟。用培养基悬浮并清洗细胞，换用新鲜培养液重悬细胞，并按照常规方法铺板。
