Tyrosyl-DNA phosphodiesterase 2 (TDP2), also known as TRAF and TNF receptor-associated protein (TTRAP), is a multifunctional enzyme critical for repairing DNA damage caused by topoisomerase II (TOP2) activity. TDP2 specifically cleaves 5'-phosphotyrosine bonds left on DNA after TOP2-mediated cleavage, resolving abortive TOP2-DNA complexes and preventing genomic instability.
