SDHB (succinate dehydrogenase complex iron sulfur subunit B) is a key component of mitochondrial complex II, which plays a dual role in the tricarboxylic acid (TCA) cycle and electron transport chain. As a tumor suppressor, SDHB is encoded by the SDHB gene located on chromosome 1p36. Mutations in this gene are linked to hereditary paraganglioma-pheochromocytoma syndrome (HPPS) and other cancers, including renal cell carcinoma and gastrointestinal stromal tumors (GISTs). SDHB deficiency disrupts cellular metabolism, leading to pseudohypoxic drive and tumorigenesis.

SDHB antibodies are widely used in immunohistochemistry (IHC) to assess SDHB protein expression in tumor tissues. Loss of SDHB immunoreactivity serves as a surrogate marker for SDHx mutations (SDHA/B/C/D), aiding in the diagnosis of SDH-deficient neoplasms. These antibodies help differentiate SDH-deficient tumors from sporadic counterparts, guiding genetic testing and clinical management. Additionally, SDHB IHC is critical in classifying GISTs, as SDHB-negative cases often exhibit wild-type KIT/PDGFRA mutations and unique clinical behavior. Research applications include studying mitochondrial dysfunction, metabolic reprogramming in cancers, and exploring therapeutic targets linked to SDH complex abnormalities. SDHB antibodies thus hold diagnostic, prognostic, and research significance in precision oncology.
