The KIR2DL3 antibody is a reagent used to study the human killer-cell immunoglobulin-like receptor 2DL3 (KIR2DL3), a member of the KIR family encoded by the leukocyte receptor complex on chromosome 19. KIRs are expressed on natural killer (NK) cells and a subset of T lymphocytes, playing critical roles in regulating immune responses by interacting with human leukocyte antigen (HLA) class I molecules. KIR2DL3 is an inhibitory receptor characterized by two extracellular immunoglobulin-like domains and a long cytoplasmic tail containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs). It specifically binds to HLA-C antigens belonging to the "C1" group (e.g., HLA-Cw*07), transmitting inhibitory signals that dampen NK cell cytotoxicity and cytokine production when self-HLA is recognized.

This receptor-ligand interaction is pivotal in immune tolerance, viral defense, and pregnancy, where KIR2DL3-HLA-C combinations influence outcomes in infections (e.g., hepatitis C clearance), reproductive disorders (e.g., preeclampsia), and cancer immunity. Antibodies targeting KIR2DL3 are essential tools for functional studies, flow cytometry profiling, and therapeutic development. Research has explored blocking KIR2DL3 to enhance NK-mediated tumor lysis in hematologic malignancies, while population studies link KIR2DL3 polymorphisms to autoimmune and infectious disease susceptibility. Its dual role in immune surveillance and pathogenesis continues to drive interest in diagnostic and immunotherapeutic applications.
