The SORT1 antibody targets sortilin, a protein encoded by the SORT1 gene, which belongs to the VPS10P-domain receptor family. Sortilin is a type-1 transmembrane glycoprotein involved in intracellular protein sorting, trafficking, and signaling. It functions as a receptor or co-receptor in various pathways, including neurotrophin signaling, lipid metabolism, and apoptosis. Structurally, it contains a luminal VPS10P domain for ligand binding, a transmembrane region, and a short cytoplasmic tail mediating endocytic sorting.

SORT1 has gained attention for its dual roles in physiological and pathological processes. In the nervous system, it regulates neurotrophic signaling by trafficking proteins like proNGF and brain-derived neurotrophic factor (BDNF), implicating it in neurodegenerative diseases such as Alzheimer’s and Parkinson’s. In cardiovascular and metabolic contexts, SORT1 modulates LDL cholesterol levels by interacting with apolipoproteins, linking it to atherosclerosis and diabetes. Additionally, it participates in proapoptotic pathways in cancer.

SORT1 antibodies are essential tools for studying these mechanisms. They enable detection, localization, and functional analysis of sortilin in tissues or cells via techniques like Western blot, immunohistochemistry, and flow cytometry. Researchers also use SORT1 antibodies to explore therapeutic strategies targeting sortilin-associated pathways, offering potential for treating neurodegenerative, cardiovascular, and metabolic disorders.
