名称	Dimethyl malonate
描述	Dimethyl Malonate is a Succinate Dehydrogenase inhibitor.
动物实验	Rats were subjected to a 6?min of CA, and cardiopulmonary resuscitation (CPR) was performed with administration of normal saline or dimethyl malonate (DMM, a competitive inhibitor of SDH). After the return of spontaneous circulation, neurological function of the rats was assessed by a tape removal test for 3?days. The rats were then sacrificed, and their brains were used to assess neuronal apoptosis by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. Hippocampal tissues were used for Western blotting analysis and biochemical detection. In addition, hippocampal mitochondria during CA and CPR were isolated. The relative mitochondrial membrane potential (MMP) and cytochrome C in the cytosol were detected.
体内活性	Dimethyl malonate抑制琥珀酸脱氢酶，减轻心脏骤停大鼠模型中的脑损伤。Dimethyl malonate抑制caspase-3的激活，增强HIF-1α的表达。此外，Dimethyl malonate抑制了CPR后过度的MMP超极化，并阻止了细胞色素C的释放。因此，通过Dimethyl malonate抑制SDH减轻了CA后的脑损伤，主要机制包括抑制MMP的过度超极化，减少mtROS的生成，并稳定HIF-1α的结构。
