名称	Acarbose
描述	Acarbose (BAY g 5421) is a hypoglycemic agent that is an inhibitor of orally active alpha-glucosidase (IC50=11 nM). Acarbose enhances the hypoglycemic effect of sulfonylureas or insulin.
细胞实验	Cell viability is determined using the MTT assay. Cells are seeded in 24-well culture plates at a density of 2×104 cells/well, incubated for 48 h, treated with acarbose at varying concentrations (0.5, 1.0, 2.0, 3.0, and 5.0 μM) for 24 h; or pre-treated with TNF-α (20 ng/mL) for either 24 h or 48 h to evaluate the dose-dependent effects of acarbose on VSMC growth and viability, cultured with 0.5 mg/mL MTT at 37°C in a humidified atmosphere of 5% CO2 for another 4 h, and solubilized with isopropanol. The viable cell number varies directly with the concentration of formazan product measured spectrophotometrically at 563 nm.
体外活性	方法： TNF-α 预处理的A7r5 细胞用Acarbose（1、2 和 3 μM）处理后使用Western Blot检测靶蛋白表达水平。结果： Acarbose剂量依赖性地降低 β-半乳糖苷酶、Ras 表达并增加 p-AMPK 表达。[1]
体内活性	方法：为研究Acarbose的降血糖作用，将Acarbose（30、60 mg/kg）灌胃给大鼠，每天一次，持续8周。结果：Acarbose降低空腹血糖，并调节 DM 大鼠的葡萄糖耐量而不减轻体重。Acarbose 显著抑制 DM 大鼠血清 IL6 和 TNF-α。[2] 方法：为研究Acarbose对动脉粥样硬化的影响，将Acarbose（2.5-5.0 mg/kg）喂食给兔子，持续25周。结果：Acarbose 显著且剂量依赖性地降低新内膜 IL-6 的强度，TNF-α 和 iNOS 染色，并显著增加新内膜 p-AMPK 染色的强度。Acarbose 显著且剂量依赖性地降低 HCD 喂养兔的新内膜 Ras 和 β-半乳糖苷酶表达，而体重没有减轻。[1]
