名称	Acyclovir
描述	Acyclovir (Aciclovir) is a guanine analog and orally active antiviral agent characterized by a narrow antiviral spectrum, high selectivity, and low toxicity. Acyclovir exhibits activity against HSV-1 (IC50 = 0.85 μM), HSV-2 (IC50 = 0.86 μM), and varicella-zoster virus. Acyclovir can be used for herpesvirus treatment research.
激酶实验	Total AMPK activity is measured using the method of Dagher et al. AMPK activity is quantified in the resuspended pellet as incorporation of?32P from [γ-32P]ATP (10 GBq/mmol) into a synthetic peptide with the specific target sequence for AMPK, the SAMS peptide. Radioactivity is measured using a liquid scintillation counter. Protein content in the solution containing the resupended (NH4)2SO4 pellet is determined using the Bradford method.
体外活性	方法：流式细胞术检测 10、100μM Acyclovir 处理 Jurkat 细胞 24、48、72h 的细胞周期分布。结果：Acyclovir 诱导 Jurkat 细胞发生 S 期周期阻滞，72h 时 sub-G1 凋亡峰显著升高。[4] 方法：台盼蓝染色法检测 3~100μM Acyclovir 处理 Jurkat、U937、K562 白血病细胞 24、48、72h 后的细胞活力。结果：Acyclovir 呈剂量 - 时间依赖性抑制 Jurkat 细胞活力，对 U937、K562 细胞的抑制作用较弱。[4]
体内活性	方法：雌性丹麦长白猪 (n=6，体重 37–48 kg)，单次静脉推注 Acyclovir (10 mg/kg)，给药后监测 8 h，每 30 分钟采样一次。结果：血浆药物浓度迅速达峰，但 CNS 分布缓慢且有限。[1] 方法：4 只健康亚洲象幼象单次静脉推注 Acyclovir (15 mg/kg)，给药前及给药后 0， 0.25， 0.5， 0.75， 1， 1.5， 2， 3， 4， 5， 6， 12， 24， 36， 48 h 采血，LC-MS/MS 检测血浆药物浓度。结果：给药后 12 小时，血浆浓度仍高于 HSV-1/2 和 EHV-1 的 IC50。[2] 方法：雄性 Sprague-Dawley 大鼠硬膜外注射 Acyclovir (0.3 mg， 0.6 mg， 0.9 mg) (溶于 100 μL)或静脉注射 Acyclovir (3 mg， 6 mg， 9 mg )(溶于 2 mL 生理盐水)，给药后 1 小时采集血液和脑脊液样本。结果：硬膜外组脑脊液 CSF 浓度显著高于静脉组 (P<0.05)，而血浆浓度显著低于静脉组 (P<0.05)。[3]
