名称	γ-Aminobutyric acid
描述	γ-Aminobutyric acid belongs to natural products and functions as an agonist of GABAA and GABAB receptors, possessing central sedative effects, cell permeability, and the ability to modulate neuronal excitability. This compound is used in neuroscience research and exhibits anxiolytic, anticonvulsant, and neuroprotective activities.
细胞实验	GABRP-positive cell lines, KLM-1 and PK-45P, and GABRP-negative cell lines, PK-59 and KP-1N, are incubated with GABA or GABA receptor agonist Muscimol at serial concentration (0, 1, 10, 100 μmol/L) in appropriate medium supplemented with 1% FBS for 6 days. To inhibit the GABA-mediated pathway, cells are incubated with 250 μmol/L of GABAA receptor antagonist bicuculline methiodide or 1 mmol/L of GABAB receptor antagonist CGP-35348. After 6 days of exposure to either of these drugs, cell viability is measured by MTT assay as described above.(Only for Reference)
体外活性	方法：GABRP阳性胰腺癌细胞（KLM-1、PK-45P）及外源性表达GABRP的HEK293细胞经γ-Aminobutyric acid (GABA)（1–100 μM）处理，并加入GABAA受体拮抗剂干预，采用MTT法、BrdU掺入法检测增殖，Fura-2测胞内Ca²⁺，Western blot检测Erk磷酸化。结果：γ-Aminobutyric acid (GABA)剂量依赖性促进细胞增殖，升高胞内Ca²⁺并激活Erk通路，该效应被GABAA受体拮抗剂抑制。[1] 方法：INS-1细胞及小鼠原代胰岛β细胞经γ-Aminobutyric acid (GABA)（100 μM）处理，采用膜片钳记录膜电位，Fura-2钙成像检测Ca²⁺内流，Western blot检测Akt磷酸化。结果：γ-Aminobutyric acid (GABA)诱导β细胞膜去极化及Ca²⁺内流，激活PI3K/Akt通路，发挥促生存及抗糖尿病效应。[2]
体内活性	方法：MDSD及NOD糖尿病小鼠每日腹腔注射GABA（20 μmol/只），连续数周，采用血糖仪监测血糖，RIA检测循环胰岛素和胰高血糖素水平。结果：γ-Aminobutyric acid (GABA)处理提高小鼠循环胰岛素、降低胰高血糖素，维持接近正常血糖水平，显著改善代谢状态。[2] 方法：SJL/J小鼠免疫PLP 139-151诱导EAE模型，每日口服GABA能药物托吡酯（100 mg/kg）或氨己烯酸（400 mg/kg），溶于PBS连续给药37天或自疾病高峰期开始治疗。ELISA检测细胞因子，H&E染色评估脊髓炎症。结果：GABA能药物显著减轻EAE临床评分，抑制巨噬细胞IL-1β、IL-6及T细胞IFN-γ、IL-17产生，减少中枢炎症浸润。[3]
