描述	Mobocertinib is a selective EGFR/HER2 inhibitor that also functions as a potent dual inhibitor of the ABCB1 and ABCG2 efflux transporters. It demonstrates high binding affinity to both proteins, effectively reversing multidrug resistance by blocking drug efflux and restoring intracellular accumulation of substrate agents. Notably, mobocertinib inhibits the ATPase activity of ABCB1 and ABCG2 without altering their expression or cellular localization. In xenograft models, mobocertinib significantly enhances the antitumor efficacy of co-administered substrate drugs, such as paclitaxel and topotecan, leading to substantial tumor regression. This compound serves as a valuable tool for investigating mechanisms of chemoresistance reversal and for repurposing studies targeting multidrug-resistant cancer models.
