In vitro studies:
Tirofiban(MK-383,Aggrastat) is a non-peptide derivative of tyrosine that selectively inhibits GP-IIb/IIIa receptors and has the lowest efficacy against alpha vβ3Vitronectin receptors. Tirofiban inhibited gel-filtered platelet aggregation induced by 10μMADP with an IC50 of 9nM, but inhibited human umbilical vein adhesion to Vitronectin with an IC50 of 62μmol/L, which was dependent on the Alpha vβ3 receptor.
In vivo studies:
Tirofiban (10 to 500mg/kg or 10micrograms/kg/min continuous intravenous infusion for 360 minutes) inhibited platelet aggregation with ADP and collagen in canine models. Tirofiban treated humans at 0.15μg/kg/min for 4 hours increased bleeding time by 2.5 times and inhibited ADP-induced platelet aggregation by 97%.
