Colistin sulfate, also known as colistin sulfate, Christian (Colistin), PolymyxinE E(PolymyxinE), antienemy, etc., white or near-white powder, odorless, bitter taste with moisture induction, easily soluble in water, slightly soluble in methanol, ethanol, almost insoluble in acetone, ether, etc., free alkali slightly soluble in water. Stable in the PH3-7.5 range. Colistin sulfate is produced by Bacillus polymyxis and has strong antibacterial effect on gram-negative bacteria. It is used to treat intestinal diseases caused by gram-negative bacteria. It is also used as feed additive and has obvious growth-promoting effect. The combination of sulfadiazine effect is better.
Pharmacological activity
The bactericidal activity of colistin sulfate is mainly due to its molecular structure of fatty acids, positively charged amino acids and cyclic peptides three key active group structure. The lack or change of any of the above structures will greatly reduce its bactericidal activity. Colistin sulfate plays a bactericidal role. It is mainly a positively charged free amino group, which can combine with the negatively charged phosphoric acid in the phospholipid in the serous membrane of bacterial cells, reducing the surface tension of the serous membrane of bacteria, destroying the integrity of the cell membrane, increasing the permeability of the cell membrane, and allowing the main components in the cell, especially purine and pyrimidine, to flow out of the cytoplasm. As a result, bacterial growth is inhibited or bacteria die. Because the cell membrane of Gram-negative bacteria contains more phospholipids, and the content of phospholipids in the cell membrane of Gram-positive bacteria is very small, so the antibacterial effect of colistin sulfate is mainly the strongest against Gram-negative bacteria. In view of this, the antimicrobial spectrum of colistin sulfate is limited to E. coli, Salmonella, Pasteurella, pseudomonas aeruginosa, Haemophilus and other gram-negative bacteria. In addition, the resistance of these bacteria to commonly used aminoglycosides, penicillins, cephalosporins and fluoroquinolones is quite common. Also, many bacteria are multidrug-resistant. However, colistin sulfate is not easy to develop resistance, so far no resistance to colistin sulfate resistant plasmid transmission has been found. Therefore, the spread and spread of resistance to colistin sulfate is very slow, and most gram-negative bacteria are sensitive to colistin sulfate.
peculiarity
(1) Has a strong ability to resist Gram-negative bacteria. In particular, it has specific developmental retarding effect on Escherichia coli, Salmonella and Pseudomonas aeruginosa.
(2) It has the effect of killing bacteria. By blocking the membrane's selective permeability function, bacteria die
(3) There is almost no problem of drug resistance. No R factor resistance was found.
(4) Synergistic effect with anti-gram-positive bacteria drugs. Combined with bacitracin zinc, flavomycin, sulfonamides, semi-synthetic penicillin, gentamicin and so on, the effect is even better.
(5) No residue. When orally administered, it is almost not absorbed by the intestine, but when intramuscular injection, blood absorption is good, so there is no need to worry about residues in livestock products.
(6) Promote the growth of livestock and poultry, and prevent infectious enteritis of livestock and poultry.
Application:
1 is a class of antibiotics, used to treat intestinal diseases caused by gram-negative bacilli, and has a certain growth promotion effect
2 Mainly used for sensitive bacterial infections, such as sepsis, acute enteritis, urinary tract infection, etc
3 Antibiotics. This product is produced by Bacillus polymyxis and has strong antibacterial effect on gram-negative bacteria. It is used to treat intestinal diseases caused by gram-negative bacteria. Colistin sulfate is used to penetrate bacterial cell membranes and mannose anticoagulants and in the study of antibiotic resistance in some organisms, such as Bowman. It has also been used to study renal hephrotoxicity, MIC value to Pseudomonas aeruginosa, temporal kinetics, and post-antibiotic effect (PAE) in rats.
