Chemical properties:

Production method:
1.5g(8mmo1) N-methyl-4-nitrophenethylamine (I) and 1.55g(7.7mmo1)2-[4-nitrophenoxy] ebromine (II) were dissolved in 50ml acetonitrile, 1.25g potassium carbonate and 1.2g sodium iodide were added, and stirred back for 72h. As for concentration, the remaining oily solids were divided between 100ml2mol/L sodium bicarbonate aqueous solution and 100ml ethyl acetate.
The water layer was then extracted with 50ml ethyl acetate for two times, the ethyl acetate layer was combined, washed with saturated sodium chloride aqueous solution, and anhydrous magnesium sulfate was dried, filtered and concentrated.
The 1.7g compound (Ⅲ) was obtained by ethanol crystallization with yield of 64% and melting point of 74℃. Compound (Ⅲ)(1.5g, 4.5mmo1) was dissolved in 100ml ethanol, catalyzed by Raney nickel and stirred at room temperature for 16h at 0.303MPa hydrogen pressure. Strain and concentrate the filtrate until dry. The remaining oil is dissolved in ether, filtered, and concentrated. Then, 0.90g compound (Ⅳ) was obtained by recrystallization with 1:1 ethyl acetate and petroleum ether mixture (60-80 ℃) with yield of 72% and melting point of 73-74 ℃. Compound (Ⅳ)(0.75g, 2.6mmo1) and mesylate anhydride (1.0g, 4mmo1) were dissolved in 50ml dry dichloromethane and stirred overnight at room temperature.
The remaining oil was divided between 50ml2mol/L sodium bicarbonate aqueous solution and 50ml ethyl acetate. The water layer was then extracted with ethyl acetate (2×25m1), the ethyl acetate layer was combined, and the anhydrous magnesium sulfate was dried, filtered, and concentrated. The remaining solid was crystallized with ethyl acetate and methanol (10:1) to obtain 0.6g dofilide with a yield of 51% and a melting point of 147 ~ 149℃.
