Produced in rabbits immunized with purified, recombinant Human IL6 / IL-6 (rh IL6 / IL-6; Catalog#10395-HNAE; NP_000591.1; Val30-Met212). IL6 / IL-6 specific IgG was purified by Human IL6 / IL-6 affinity chromatography.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-IL-6 antibody (ab6672)
IHC image of IL6 staining in human lung formalin fixed paraffin embedded tissue section*, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab6672, 1/400, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Interleukin-6 (IL-6) is a multifunctional α-helical cytokine that regulates cell growth and differentiation of various tissues, which is known particularly for its role in the immune response and acute phase reactions. IL-6 protein is secreted by a variety of cell types including T cells and macrophages as phosphorylated and variably glycosylated molecule. It exerts actions through the its heterodimeric receptor composed of IL-6R that lacks the tyrosine/kinase domain and binds IL-6 with low affinity, and ubiquitously expressed glycoprotein 13 (gp13) that binds the IL-6. IL-6R complex with high affinity and thus transduces signals. IL-6 is also involved in hematopoiesis, bone metabolism, and cancer progression, and has been defined an essential role in directing transition from innate to acquired immunity.
双抗体夹心法测定标本中HUMAN IL-6水平。用纯化的HUMAN IL-6抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入HUMAN IL-6，再与HRP标记的HUMAN IL-6抗体结合，形成抗体-抗原-酶标抗体复合物，经过彻底洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的HUMAN IL-6呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），通过标准曲线计算样品中HUMAN IL-6浓度。
ANTI-HUMAN IL-6是一类可以特异性结合HUMAN IL-6的多克隆抗体，主要用于检测HUMAN IL-6的Western Blot、IHC-P、IF、ELISA、Co-IP等多种免疫学实验。
白细胞介素-6(Interleukin-6)，简称白介素6(IL-6)，是一种功能广泛的多效性细胞因子。IL-6可调节多种细胞的生长与分化，具有调节免疫应答、急性期反应及造血功能，并在机体的抗感染免疫反应中起重要作用。
IL-6在多种疾病时有明显改变，其表达失调可引起许多疾病，其临床表现主要为发病时IL-6水平增高。在发生内外伤、外科手术，应激反应，感染，脑死亡，肿瘤产生以及其它情况的急性炎症反应过程中IL-6会快速生成。手术病人的IL-6浓度能够预示是否会有手术并发症的产生。
连续检测重症监护病人血清或血浆中IL-6的水平能有效的评估系统性炎症反应综合症（SIRS）的严重程度，脓毒血症以及脓毒血症性休克的预后情况。IL-6的血清浓度水平增高早于PCT和CRP，能作为脓毒血症的早期警告指标。IL-6还在慢性炎症反应（如类风湿关节炎）中扮演着重要角色。
