以2-溴-6-氟吡啶和联硼酸频那醇酯为原料合成6-氟-2-吡啶硼酸频哪醇酯的一般步骤:在氮气保护下,将Pd(dppf)2Cl2·CH2Cl2(102 mg,0.14 mmol)加入到脱气的2-溴-6-氟吡啶(410 mg,2.33 mmol)、双(频哪醇合)二硼(828 mg,3.26 mmol)和KOAc(685 mg,6.99 mmol)的二恶烷(6 mL)溶液中。反应混合物在115℃下搅拌加热1小时。反应完成后,冷却至室温,通过减压蒸馏除去溶剂。残余物用硅胶柱色谱法纯化(洗脱剂:二氯甲烷/甲醇,100:0至90:10),收集含目标产物的馏分,减压浓缩后得到6-氟-2-吡啶硼酸频哪醇酯(400 mg,收率76%)。产物结构经1H NMR(400 MHz,CDCl3)确认:δ 7.78(td,J = 8.1, 7.2 Hz,1H),7.70(ddd,J = 6.9, 2.8, 0.9 Hz,1H),6.98(ddd,J = 8.1, 2.7, 0.9 Hz,1H),1.38(s,12H)。
