ChEBI: O-phospho-L-threonine is a L-threonine derivative phosphorylated at the side-chain hydroxy function. It has a role as an Escherichia coli metabolite. It is a L-threonine derivative, a non-proteinogenic L-alpha-amino acid and an O-phosphoamino acid. It is a conjugate acid of an O-phosphonato-L-threonine(2-).
Dissolve the phosphate in the minimum volume of H2O, add charcoal, stir for a few minutes, filter and apply onto a column of Dowex 50W (H+ form), then elute with 2N HCl. Evaporate the eluates under reduced pressure whereby the desired fraction produces crystals of the phosphate which can be recrystallised from H2O/MeOH mixtures and the crystals are then dried in vacuo over P2O5 at ~80o.
